- María Mexía Bocanegra, the wife of Diego Gutiérrez de la Caballería
- Isabel Ortiz, the wife of Cristóbal de Escobar.
- Doña Luisa, the wife of Jorge Báez, may have been a mestiza woman.
- Beatriz Gutierrez, the wife of Juan Hernández.
- Isabel was the wife of Francisco Martín. Historical records indicate that Isabel was an American Indian woman.
- Doña Maria de Zárate was the niece of the bishop of Oaxaca and wife of Juan Gallego.
- Catalina Mejía de la Torre was the daughter of Melchior Pérez and wife of Pedro de Ledesma. Pedro de Ledesma and Catalina Mejía de la Torre married following Pérez's service on the expedition.
- Leonor Padilla was the daughter of Cristóbal Romero (a conquistador with Cortés) and the wife of Francisco de Olivares.
- Marina de Robles was the wife of Cristóbal de Quesada. Though Quesada relocated to New Spain following the expedition, historical records indicate that Marina had not joined him as late as 1547.
- Doña Francisca de Guzmán was the daughter of the governor of Cuba. She was married to don Pedro de Tovar.
- Beatriz de Estrada, daughter of Alonso de Estrada, the royal treasurer of New Spain, and doña Marina Gutiérrez de la Caballería, was married to Francisco Vázquez de Coronado.
- Francisca de Hozes, wife of Alonso Sánchez, was an important witness in the 1544 investigation of the expedition's treatment of Native Americans
- María Maldonado married Juan Gómez de Paradinas just before the expedition began. She nursed sick and injured expeditionaries.
- María Zambrano was the wife of Juan Martín.
- Elena Bonilla was Cristóbal de Mayorga's wife. He predeceased her.
- Doña Catalina de Santa Gadea was the wife of Pablo de Melgosa. She never left Spain, and he returned there after the expedition.
- Doña Catalina de Sotomayor married Pero Méndez de Sotomayor after her first husband died.
- Elvira de Salcedo was the first wife of Gerónimo Mercado de Sotomayor. He subsequently married doña Catalina de Polanco.
- After Antón Miguel died early in 1543, his wife whose name is not known remarried to Pascual Bernal.
- Francisca de la Torre was the first wife of Andrés de Miranda, who then married a daughter of Diego de Ortuño.
- Juana de Torija was married to Alonso del Moral.
- María de Arriaga was the wife of Francisco Muñoz.
- Pedro de Castañeda de Nájera was married, but his wife's name is not known.
- Catalina López was married to Pedro Navarro.
- Antón Negrín was married to an unknown Native American woman.
- Francisco de Olivares was the husband of Leonor Padilla.
- María González was married to Sancho Ordóñez.
- Andrés Orejón had a wife in Spain whose name is not known.
- A María López was the wife of Fernán Páez.
